Protocol

  1. Cool cells if needed, protocol also works at 37C
  2. Wash 2X with PBS at required temperature
  3. Cover cells with PEG media, incubate 10-20 min
  4. Wash carefully with PBS to remove PEG.

Recipes

1L PEG Media:
  • 4.5g Sucrose
  • 1.907g 5mM EGTA
  • 1L PBS
10ml PEG Media:
  • 0.045g Sucrose
  • 0.019g 5mM EGTA
  • 10ml PBS
Warning: cells will be weakly adherent after this procedure; take care to minimize shear in order to avoid loosing the cells.
